The 33-year-old’s contract was expected to leave the club with his contract expiring this summer.
Vertonghen made a total of 315 appearances for the North London club, scoring 12 goals.
Though he never won a trophy with the club, he was part of the Tottenham side that finished in the top four consecutively and also made it to the Champions League final.
Vertonghen joined Tottenham from Ajax in 2012.
It remains unclear where the Belgian defender will play next season.
Spurs are likely to be active in the transfer market this summer as they seek a replacement for Vertonghen.
Jose Mourinho’s side finished 6th in the Premier League after a 1-1 draw on Sunday at Crystal Palace.
